組成結(jié)構(gòu)
1、血清：操作過程中避免任何細(xì)胞刺激.使用不含熱原和內(nèi)毒素的試管.收集血液后,1000×g離心10分鐘將血紅細(xì)胞迅速小心地分離.
2、血漿：EDTA、檸檬酸鹽、肝素血漿可用于檢測(cè).1000×g離心30分鐘去除顆粒.

4、組織勻漿：將組織加入適量生理鹽水搗碎.1000×g離心10分鐘,取上清液.

操作方法
雙抗體夾心法
1.包被：用0.05MPH9.牰碳酸鹽包被緩沖液將抗體稀釋蛋白質(zhì)含量為1～10μg/ml.在每個(gè)聚苯乙烯板的反應(yīng)孔中加0.1ml,4℃隔夜.次日,棄去孔內(nèi)溶液,用洗滌緩沖液洗3次,每次3分鐘.（簡(jiǎn)稱洗滌,下同）.
2. 加樣：加一定稀釋的待檢樣品0.1ml于上述已包被之反應(yīng)孔中,置37℃孵育1小時(shí).然后洗滌.（同時(shí)做空白孔,陰性對(duì)照孔及陽(yáng)性對(duì)照孔）.
3. 加酶標(biāo)抗體：于各反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)抗體（經(jīng)滴定后的稀釋度）0.1ml.37℃孵育0.5～1小時(shí),洗滌.
4. 加底物液顯色：于各反應(yīng)孔中加入臨時(shí)配制的TMB底物溶液0.1ml,37℃10～30分鐘.
5. 終止反應(yīng)：于各反應(yīng)孔中加入2M硫酸0.05ml.
6. 結(jié)果判定：可于白色背景上,直接用肉眼觀察結(jié)果：反應(yīng)孔內(nèi)顏色越深,陽(yáng)性程度越強(qiáng),陰性反應(yīng)為無色或極淺,依據(jù)所呈顏色的深淺,以“+"、“-"號(hào)表示.也可測(cè)OD值：在ELISA檢測(cè)儀上,于450nm（若以ABTS顯色,則410nm）處,以空白對(duì)照孔調(diào)零后測(cè)各孔OD值,若大于規(guī)定的陰性對(duì)照OD值的2.1倍,即為陽(yáng)性.
間接法
1.用包被緩沖液將已知抗原稀釋1～10μg/ml,每孔加0.1ml,4℃隔夜；
2.次日洗滌3次；
3.加一定稀釋的待檢樣品（未知抗體）0.1ml于上述已包被之反應(yīng)孔中,置37℃孵育1小時(shí),洗滌；
4.（同時(shí)做空白、陰性及陽(yáng)性孔對(duì)照）于反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)第二抗體（抗抗體）0.1ml；
5.37℃孵育35-60分鐘,洗滌；
6.’zui后一遍用DDW洗滌.
其余步驟同“雙抗體夾心法"的4、5、6.
