elisa技術(shù)流程
ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標(biāo)記.結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學(xué)活性,酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性,又保留酶的活性.在測定時(shí),受檢標(biāo)本（測定其中的抗體或抗原）與固相載體表面的抗原或抗體起反應(yīng).用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與液體中的其他物質(zhì)分開.再加入酶標(biāo)記的抗原或抗體,也通過反應(yīng)而結(jié)合在固相載體上.此時(shí)固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例.加入酶反應(yīng)的底物后,底物被酶催化成為有色產(chǎn)物,產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān),故可根據(jù)呈色的深淺進(jìn)行定性或定量分析.由于酶的催化效率很高,間接地放大了免疫反應(yīng)的結(jié)果,使測定方法達(dá)到很高的敏感度.ELISA可用于測定抗原,也可用于測定抗體.
然而,影響Elisa試驗(yàn)結(jié)果的因素很多,故加強(qiáng)各個(gè)環(huán)節(jié)的質(zhì)量保證才能充分發(fā)揮其方法學(xué)的優(yōu)點(diǎn).
試驗(yàn)原理
試劑盒是固相夾心法酶聯(lián)免疫吸附實(shí)驗(yàn)（ELISA）.已知濃度的標(biāo)準(zhǔn)品、未知濃度的樣品加入微孔酶標(biāo)板內(nèi)進(jìn)行檢測.先將生物素標(biāo)記的抗體同時(shí)溫育.洗滌后,加入親和素標(biāo)記過的HRP.再經(jīng)過溫育和洗滌,去除未結(jié)合的酶結(jié)合物,然后加入底物A、B,和酶結(jié)合物同時(shí)作用.產(chǎn)生顏色.顏色的深淺和樣品中的濃度呈比例關(guān)系.
原理
免疫測定技術(shù)的基礎(chǔ)在于抗原抗體之間的特異結(jié)合反應(yīng),所以任何優(yōu)質(zhì)的診斷試劑離不開優(yōu)質(zhì)的原料,如抗原抗體,酶等.以前用于免疫測定的抗原通常為各種純化抗原,而抗體則為純化抗原免疫動(dòng)物后獲得的多克隆抗體和使用雜交瘤技術(shù)得到的單克隆抗體,而抗原或抗體的標(biāo)記物如酶標(biāo)結(jié)合物則通過各種化學(xué)合成方法制備.近年來,隨著分子生物學(xué)的發(fā)展,使用基因工程方法制備各種特殊的抗原或抗體及其酶結(jié)合物等免疫測定試劑幾成為現(xiàn)實(shí)的新一代試劑,各種新型使用的測定方法也不斷出現(xiàn).
HBsAg試劑特點(diǎn)（檢測模式：臨床抗體夾心法）：包被抗體為山羊多抗.多抗具有高親和性和對抗原各種表位的反應(yīng)性.酶表抗體為與HBsAg有不同結(jié)合位點(diǎn)的鼠復(fù)合單位,可測得HBsAg變異樣品,提高檢測的特異性（99.98%）提高檢測的靈敏度,應(yīng)用ParlEhrich學(xué)說（PEI）HBsAg標(biāo)準(zhǔn)品,對ad標(biāo)準(zhǔn)品的靈敏度為0.05ng/ml對ay標(biāo)準(zhǔn)品靈敏度為0.025ng/ml
ELISA檢測試劑盒應(yīng)用定性夾心免疫檢測技術(shù),用合成的HEV多肽抗原包被微孔板板條,這些多肽是中國型HEV毒株核心氨基酸序列中抗原性很強(qiáng)的肽段,分別來自于該毒株的開放閱讀框2和開放閱讀框3.將樣品或標(biāo)準(zhǔn)品加入孔中并孵育,如果其中存在HEVIgM抗體,這些抗體就會(huì)與HEV的多肽抗原結(jié)合,并固定在上面,洗板除去其它非特異性抗體和樣品中的其它成份.然后加入羊抗人IgM-HRP（辣根過氧化物酶）酶結(jié)合物,經(jīng)第二次孵育后,酶結(jié)合物就會(huì)與*次孵育結(jié)合上的HEVIgM抗體相結(jié)合,洗板除去未結(jié)合的酶結(jié)合物,加入TMB底物溶液,在第三次孵育時(shí)會(huì)發(fā)生酶-底物反應(yīng),只有那些含有HEVIgM抗體和酶結(jié)合物所形成的復(fù)合物的孔才會(huì)發(fā)生顏色變化,加入硫酸溶液終止酶和底物間的反應(yīng),并在波長:450nm處測量O.D.值,按照本HEVIgM抗體elisa試劑盒的測試標(biāo)準(zhǔn),O.D.值大于或等于Cut-Off值的樣品被認(rèn)為是初試陽性.
